The effect of scopoletin and rutin on the reinforcing properties of ethanol using conditioned place preference test in mice

Abstract:

The conditioned place preference (CPP) test is widely used to study the rewarding properties of many addictive substances, including ethanol. In the present study, the therapeutic potential of scopoletin and rutin against the rewarding properties of ethanol using a mouse CPP test was investigated. The CPP methodology comprised of: habituation (3 days), preconditioning (1 day), conditioning (10 days), rest (1 day) and post-conditioning (1 day). In the post-conditioning day, vehicle-saline control and vehicle-ethanol control group received 1% w/v sodium carboxymethyl cellulose solution (CMC) (10 ml/kg body weight (bw), p.o.); scopoletin, rutin and acamprosate -treated groups received the respective drugs orally 1 h prior to the preference test. Vehicle-ethanol control group showed a marked preference to the ethanol-paired white compartment of the CPP box on the postconditioning day (day 16). Interestingly, the acute oral administration of scopoletin and rutin, 1 h prior to testing on day 16 resulted in a reversal of preference to the ethanol-paired white compartment. The reference drug acamprosate also showed a similar outcome in the CPP test. This result discloses that scopoletin and rutin attenuated the ethanolinduced CPP in mice.

Prof Vijayapandi Pandy is hailed from Usilampatti, Madurai, Tamil Nadu and is presently working in the Department of Pharmacology, Chalapathi Institute of Pharmaceutical Sciences, Guntur, Andhra Pradesh, India. He has 20+ years of teaching and research experience in India, Malaysia, Ethiopia and Libya. He completed his PhD in one of the premier institutes of India, BITS-Pilani, Rajasthan in the year 2006. He did his M. Pharm (Pharmacology) from SCS College of Pharmacy, Harapanahalli, Karnataka in the year 2000 and B. Pharm from Arulmigu Kalasalingam College of Pharmacy (AKCP), Krishnan koil, Tamil Nadu in the year of 1997.
