Plant bioreactor has enormous system that supports many biological activities i.e., production of plant bodies, virus-like particles (VLPs) and vaccines. Foreign gene-expression is an efficient mechanism for getting protein vaccines against different human viral and non-viral diseases. Plant makes it easy to deal with, safe, inexpensive and easy storage. Broad spectrum of safe gene promoters is being used to avoid risk assessments. Engineered virus-based vector has no side effect. The process can be manipulated as a) retrieve and selection of gene encoding an antigenic protein from Gene Bank and or from a viral-genome subunit sequence isolated as local strain. b) Design and construction of hybrid-virus vector. c) Design of chimeric virus (viral vector with gene of interest) flanked by plant-specific genetic regulatory elements for constitutive expression is manipulated. d) Gene transformation and or transfection, for transient expression, into a plant-host model i.e. tobacco to getting protocols processed positively and then moving into edible host plant. e) Confirmation of protein expression by Bioassay, PCR-associated tests (RT-PCR), Northern and Western Blotting Analysis, Serological assay (ELISA). f) Expression for adjuvant recombinant protein seeking better antigenicity is recommended. g) Extraction and purification of expressed protein for identification and dosing. h) Antigenicity capability has to be evaluated using parental or oral delivery in animal model (mice and or rabbit immunization). i) Enhancement of construct-treated edible crops by: Growing construct-treated edible crops in protective green houses. Some successful cases, of heterologous gene-expressed protein as edible vaccine, are being discussed i.e. a) Hepatitis C virus (HCV). HCV-HVR-1 and HCV-R9 were used as a potential neutralizing epitope of HCV. Both mimitope and epitope were transfected using AlMV CP P3/RNA3 and TMV CP-TMGMV CP expression vectors into tobacco plants. Expressed recombinant protein has not only been confirmed as therapeutics but also as diagnostics. B) Herpes Simplex Virus 2 (HSV-2). HSV-2gD and HSV-2VP16 subunits were transfected using TMV CP-TMGMV CP expression vector into tobacco plants.
Aboul-Ata Elnady is Professor of Plant virology, who was born on Mar 22nd 1948 in Sharkia, Egypt, is the president of ArSV (Arab Society for Virology) in 2013 for 3 years. He was formerly Department Head of Plant Virus and Phytoplasma Research, Plant Pathology Research Institute at Agricultural Research Center, Giza, Egypt for 3 years (2003-2006), and Head of Molecular laboratory at Plant Virus and Phytoplasma Res. from Sep. 2005 up to now. In 2007 he has been awarded committee membership of Egyptian Society of Advanced Material and Nanotechnology. He is being a committee-member of Egyptian Society of Virology since initiation in 2001. He is being member of the Molecular Virology Group at the University of Kentucky, USA, American Phytopathological Society, The New York Academy of Sciences, American Society for Microbiology.
