Abstract:

Skin ageing is a complex process triggered by intrinsic and extrinsic factors. Peroxissome proliferator-activated receptor (PPAR) isophorms (PPARα, PPARβ/δ or PPARγ) modulates several biological functions and in skin their expression is important to skin homeostasis maintenance. Studies have shown that PPAR isophorms activation is important to regulate matrix metalloproteinase-1 (MMP-1) expression, which acts on extracellular matrix degradation and contribute to morphological and functional changes in aged skin. In this work we evaluated MMP-1 expression in human dermal fibroblasts treated with extract of endophytic fungi that were shown to activate three isophorms of PPAR, and with agonists of three isophorms of PPAR. Human dermal fibroblasts were treated for 48 hours with a non toxic concentration (10 μg.mL-1) of endophytic fungus extractor with 20 μM of agonists of PPARα, PPARβ/δ or PPARγ, or vehicle (control). Thereafter, the cells were exposed to oxidative stress by treatment with 300 μM of H2O2 for 1 hour, then plates were washed with PBS and refresh medium was added to the plates and they were incubated for 48 h. Cells were harvested to total RNA isolation. Expression of MRNA was determined by quantitative real-time PCR. The treatment with 10 μg.mL-1 of extract decreased 78.8% of MMP-1. There was a decrease of MMP-1 expression of 79.59%, 76.82% and 76.76% to cells treated with PPARα, PPARβ/δ or PPARγ, respectively. In conclusion, extract of the endophytic fungus isolated from Bauhinia variegate leaves was able to decrease MMP-1 expression in human dermal fibroblasts as well as PPAR agonists.
