H NMR-derived serum metabolomics of oral carcinoma: A probative discriminant analysis classification model

Abstract:

Oral cancer is the sixth commonest cancer worldwide with enormous mortality. If identified at an early stage, the survival rate would be improved. Hence, new biomarkers that facilitate to detect oral cancer at an early stage are potentially important. Patient?s enrolled from the department of oral pathology and microbiology, KGMU, Lucknow after the approval of institutional ethics committee. The study population comprised of male subjects aged 20-40 years with confirmed OSCC (n=33) and OLK (n=27). This study appraised the feasibility of using 1H nuclear magnetic resonance (1H NMR) based metabolomics in the identification of the discriminates in serum from patient?s suffering with oral squamous cell carcinoma (OSCC), oral leukoplakia (OLK), and the healthy control (HC). 1H NMR derived data were processed by supervised partial least squares discriminate analysis (PLS-DA) to reveal the differentiations among these groups. PLS-DA-derived model validity was appraised applying an internal cross-validation and receiver operating characteristic (ROC) examinations. PLSDA- derived serum biomarkers (threonine, glutamine, propionate, and choline) were able to accurately segregate 93% of oral cancer (OLK+OSCC) cases with substantial sensitivity and specificity when compared to HC. The combination of acetone, glutamine, acetate, and choline were sufficient to discriminate 94% of cases of OLK with considerable sensitivity and specificity when compared to OSCC. This study reveals various striking features of NMR derived metabolomics of human serum samples of oral cancer.
