Abstract:

Q uetiapine Fumarate is an Antipsychotic drug used for the treatment of schizophrenia. It is class II drug as per Biopharmaceutical classification system (BCS). Its solubility is decreasing as the pH is increasing therefore low absorption is expected at higher pH environment. It is having low oral bioavailability therefore a floating system is desired to increase the residence time in the stomach, facilitate more amount of drug to be dissolved and that further improves the bioavailability. In the present study floating drug delivery system of Quetiapine Fumarate is prepared by Hypromellose (Hydroxypropyl methyl cellulose) as a polymer with different grades (HPMC K 100 LV and HPMC K 4 M) using Sodium bicarbonate as a gas generating agent. The formulations were optimized on the basis of in vitro buoyancy lag time, total floating time, and in-vitro drug release in 0.1 N Hydrochloric acid. In-vitro drug releases were compared with marketed formulation of Seroquel? XR. The optimized formulation shows satisfactory stability at accelerated condition of 40?C/75%RH. The in-vitro release data was fitted into different mathematical equations such as zero order, first order, Higuchi square root, and Peppas equations to find out the release kinetics. From this study it can be concluded that Quetiapine Fumarate can be formulated as floating drug delivery system
