The present study focussed to investigate the cytotoxic effect of halophilic bacteria isolated from the Kelambakkam salt pan against different human cancer cell lines. Among 63 halophilic bacterial crude extracts, 2, 7 and 41 exhibited cytotoxicity on A549, HepG2 and MCF-7 human cancer cell lines, respectively at 1 mg/ml by MTT assay. The Halophilic bacterial crude extracts treated human cancer cells attained various morphologies and structural changes, which deviated from the normal human cancer cells. Based on the cytotoxicity, 12 halophilic bacterial crude extracts were shortlisted based on the IC50 value. The polyphasic taxonomical study has revealed that the selected halophilic bacterial isolate MML1909 is Halomonas pacifica. The purification of the crude extract of H. pacifica MML1909 using column chromatography resulted in 18 fractions, the fraction 14 were selected based on IC50 as importantly, the fraction 14 were not toxic to normal human vero cell line. It was further purified by column chromatography followed by preparatory TLC and pure compound was characterized by various spectral analyses such as IR, UV, 1HNMR and 13C NMR, which revealed the molecular formula as C20H30N4O6 with molecular weight as 422.48 by ESI-MS. The pure compound of H. pacifica MML1909 IC50 determined as 45 µg and 25 µg respectively. The apoptotic morphological changes in pure compound treated A549 and MCF-7 cell lines were observed by staining with acridine orange and ethidium bromide followed by flow cytometry and DNA fragmentation analyses. These results highlighted the importance of halophilic bacteria as a novel source for antiproliferative bioactive compounds.

She was a Research Officer in an ISO Organization, worked in Clinical Trials and Epidemiological Studies. She has published papers and presented posters in international conference. She had been a Study Councellor for distance education program and serving as a Women District Organiser in NGO Organization. She has been serving as an Editorial Member of the Lifestyle magazine and a member of the NABS.
