Antifungal perspective of novel 1,2,4-triazole derivatives

Abstract:

Novel ten {2-(3-R-1H-1,2,4-triazol-5-yl)phenyl}amines were synthesized and evaluated by LC-MS, 1Н NMR and elemental analysis data. Their high affinity to sterol 14α-demethylase of Candida albicans (CYP51) was calculated by means of in silico molecular docking. The in vitro inhibition tests against Aspergillus niger and common agricultural pathogens as Fusarium oxysporum, Colletotrichum higginsianum and Phytophthora infestans showed a broad spectrum of moderate to strong antifungal properties at a concentration of 50 µg/mL. Moreover, activities of two substances 6 and 9 were equal or higher than those of references Cyproconazole and Hymexazole against C. higginsianum in 50 µg/mL or 25 µg/mL. The good relationship between affinity to CYP51, lipophilicity, structure and activity was found. The obtained results demand expansion of the range of novel 1,2,4-triazole antifungal agents, investigate their activity mechanism and environmental friendliness.

Biography :

Antypenko Lyudmyla has completed her PhD of Pharmacy in Zaporizhzhya State Medical University (Ukraine, 2010). She was working from PhD student till Associate Professor at Pharmaceutical Chemistry and Organic and Bioorganic Chemistry Departments of the same university (Ukraine) till 04.2017. Afterwards till now she is working as PostDoc in project "OxiLifungi", led by Prof. Dr. Leif-Alexander Garbe in Neubrandenburg University of Applied Sciences. She has published more than 20 papers in reputed journals. Zhanar Sadykova completed her degree of Bioproduct and Food Technology in Neubrandenburg University of Applied Sciences (Germany, 2014). Afterwards she worked in the bakery industry for one year. From April 2015 till now she is scientific researcher-microbiologist in the project "OxiLiFungi" under lead of Prof. Dr. K. Steffens as a part of “Trihydroxy fatty acids as novel biological fungicides in the crop and food industries”. She is also PhD student at Applied Microbiology Department of Berlin Technical University under the supervision of Prof. Dr. Vera Meyer.
