Abstract:

Tissue culture, considered as a tool for mass multiplication especially for ornamentals was tried for tuberose cultivar Suvasini. Among the different explants tried axillary buds were found to be superior among petal segments, immature flower buds, nodal segments of rachis, stem scale segments and stem disc explants. Axillary buds when sterilized and inoculated on full MS supplemented with BAP (4mg/l) + NAA (0.5mg/l) produced multiple shoots. These multiple shoots later tried for elongation after 3rd subculture on GA3 and non GA3 supplemented MS medium. The elongated shoots later produced roots of good quality and highest number (12.75) in MS supplemented with IBA (3mg/l) + NAA (1mg/l). The well rooted plantlets later tried for acclimatization and successfully transferred to the ex vitro condition.
